Top 10 Biotech Companies To Watch In Right Now

They're demanding. They take risks -- but only calculated ones. And they ask lots of penetrating questions to understand as much as possible before putting a dime on the line. Venture capitalists, or VCs, funded nearly 3,700 deals last year. Many of those deals will fail, but the ones that don't could produce astronomical gains.

Investing in biotech stocks can be fraught with risk, but thinking like a venture capitalist can help increase the odds of success. Here are three VC principles that biotech investors should keep in mind.

1. Look for a disruptive technology that addresses an unmet need
Venture capitalists look for companies that target an unmet need and provide an innovative way to meet that need. The good news for prospective biotech investors is that there are plenty of companies fulfilling that criterion.

Perhaps the most obvious example of addressing an unmet need lies in targeting rare orphan diseases. For example, cystic fibrosis, while widely known, affects only around 30,000 Americans. Although some treatments exist to help improve qualify of life, there is no cure for the disease.

Best Warren Buffett Companies To Invest In 2015: Prothena Corporation PLC (PRTA)

Prothena Corporation PLC, incorporated on September 26, 2012, is an Ireland-based, clinical-stage biotechnology company. The Company is engaged in discovering and developing monoclonal antibodies that are directed towards misfolded proteins or improper cell adhesion. Its pipeline includes NEOD001, PRX002 and PRX003. The Company work in protein misfolding could result in therapies to treat several neurodegenerative diseases, including AL (primary) and AA (secondary) forms of amyloidosis (NEOD001), Parkinson's disease and related synucleinopathies (PRX002). Its cell adhesion development activities could generate new therapies to treat inflammatory diseases and metastatic cancers (PRX003). The Company program, NEOD001, is in Phase 1. In addition to antibodies directed to neo-epitope targets, it is developing antibodies directed to other targets. The Company has generated antibodies against cell adhesion targets expressed on certain pathogenic Th17 immune cells and tumor cells.

The Company pipeline also includes several late discovery stage programs for which it is testing antibodies in preclinical models of disease. It is also generating additional antibodies against other targets involved in protein misfolding and cell adhesion for characterization in vivo and in vitro.

NEOD001 for Amyloidosis

NEOD001 is a monoclonal antibody that targets the amyloid that accumulates in both AL and AA forms of amyloidosis. The antibody was designed to not react with normal serum amyloid A and only with the aberrant cleaved form of the protein (amyloid A).

PRX002 for Parkinson Disease

The Company has generated antibodies targeting alpha-synuclein that may slow or reduce the neurodegeneration associated with synuclein misfolding and/or transmission. It has tested these antibodies in various cellular and animal models of synuclein-related disease. In a transgenic mouse model of Parkinson disease, passive immunization with 9E4, a murine ver! sion of PRX002, reduced the appearance of synuclein pathology, protected synapses and improved performance by the mice in behavioral testing. The humanized antibody product candidate PRX002 has advanced into manufacturing and preclinical testing.

PRX003 for Inflammatory Diseases and Cancers

The Company is developing PRX003, a monoclonal antibody targeting MCAM for the potential treatment of inflammatory diseases and cancers. Still, the big challenge for Abbott will come from the medical-device arena, where many of Abbott's peers are struggling. Johnson & Johnson (NYSE: JNJ  ) has seen double-digit percentage sales declines in its cardiovascular equipment division, with growth coming solely from its major acquisition of Synthes. Meanwhile, Boston Scientific (NYSE: BSX  ) has suffered even steeper declines in sales than Abbott, as its cardiac rhythm management business in particular has seen increased pricing pressure from competition. Abbott and Boston Scientific both compete in the drug-eluting stent niche, and even though Abbott has come out with recent advances, its stent sales were down 15% in the U.S. last quarter. Still, the company sees the area as having promise, as just this morning, Abbott announced it would acquire leg-stent maker Idev for $310 million.

Johnson & Johnson (NYSE: JNJ) managed to squeeze out gains of 3.146%. What is amazing is that an analyst downgrade on Friday did not deter J&J’s resolve. Barclays cut the rating to Equal Weight from Overweight. The healthcare products and consumer products giant closed the week out at $94.74, just shy of its all-time high of $95.99. J&J’s current consensus price target is up at $98.85 as of now. J&J also offers that 2.8% dividend yield.

Johnson & Johnson (NYSE: JNJ  ) has elected to maintain its dividend. The company declared a distribution of $0.66 per share, to be handed out on Sept. 10 to shareholders of record as of Aug. 27. That amount matches the company's previous distribution, which was paid in mid-June. Before that, Johnson & Johnson's disbursement was $0.61 per share.

    An oldie, but goodie
Technically,Johnson & Johnson (NYSE: JNJ  ) isn't a pure pharma since it also sells medical devices and consumer health products, but it's that diversity that makes it such a stable dividend stock.

  • [By Dan Carroll]

Medivation (NASDAQ: MDVN  ) shares also were hit hard this week, falling 7.8% over the past five days. Shares fell 8% alone on Monday, after Johnson & Johnson (NYSE: JNJ  ) agreed to purchase Aragon Pharmaceuticals in a $1 billion deal. It wasn't a game-changer for J&J, considering Aragon's developmental prostate cancer therapy ARN-509 is in phase 2 trials, although if the company can advance the drug to a regulatory victory down the road, it could one day fill in for J&J's current blockbuster oncology drug Zytiga after its patent expires.

Xtandi improved median overall survival to 18.4 months, compared with just 13.6 months for the placebo in trials while Xofigo's median overall survival tallied 14 months, compared with 11.2 months for the control arm.

Top 10 Biotech Companies To Watch In Right Now: KYTHERA Biopharmaceuticals Inc (KYTH)

KYTHERA Biopharmaceuticals, Inc., incorporated in June 2004, is a clinical-stage biopharmaceutical company focused on the discovery, development and commercialization of prescription products for the aesthetic medicine market. The Company initial focus is on the facial aesthetics market. The Company product candidate, ATX-101, is a injectable drug in Phase III clinical development for the reduction of submental fat, which commonly presents as an undesirable double chin. Based on clinical trials conducted, ATX-101 has exhibited results in the reduction of submental fat. ATX-101 contains a synthetic form of sodium deoxycholate. In the United States and Canada, the Company is conducting two pivotal Phase III trials of ATX-101 for the reduction of submental fat. The Company initiated this pivotal Phase III clinical program, with planned enrollment of 1,000 patients, in March 2012.

In Europe, Bayer, its collaborator outside the United States and Canada, recently completed two pivotal Phase III trials of ATX-101 for the reduction of submental fat. In these multi-center, randomized, double-blind, placebo-controlled pivotal trials involving 723 patients, ATX-101 resulted in a reduction in submental fat, as assessed by a validated clinician scale and a patient satisfaction scale.

ATX-101 contains a synthetic form of sodium deoxycholate. ATX-101 is designed to be a locally-injected drug that causes proximal, preferential destruction of adipocytes, or fat cells, with minimal effect on surrounding tissue. Upon subcutaneous injection under the skin, ATX-101 disrupts cell membranes in protein-poor tissues, such as fat, while being attenuated by interactions with protein-rich tissue, such as skin, muscle and blood vessels. This attenuation by protein-rich tissue results in the preferential destruction of adipocytes by ATX-101. The destruction of adipocytes, or adipocytolysis, elicits a natural response, in which macrophages are attracted to remove cellular debris and fat particles t! hrough the lymphatic system. The macrophages also emit low levels of chemical messengers, known as cytokines, which attract fibroblasts, another cell type, to the area. Fibroblasts produce collagen, and it is believed that new collagen production, or neocollagenesis, promotes retraction of the skin in the areas of fat reduction. The fat removal process with ATX-101 is incremental with each treatment, thereby allowing for control of the aesthetic outcome.

On Tuesday, small cap biotech stock Kythera Biopharmaceuticals Inc (NASDAQ: KYTH) surged around 25% after announcing that its ATX-101 REFINE-1 and REFINE-2 Phase III trials met all primary and secondary endpoints for the reduction of so-called double chins; but if investors missed out on that rally, small caps Zeltiq Aesthetics Inc (NASDAQ: ZLTQ), Solta Medical Inc (NASDAQ: SLTM) and Cynosure, Inc (NASDAQ: CYNO) each have a piece of the aesthetic market as well. In the case of Kythera Biopharmaceuticals, its ATX-101 can be injected to deal with double chinsmeaning its less invasive than liposuction as the drug dissolves fat cells but leaves other tissue alone. JP Morgan has noted:
